During learning, the brain requires an exceptional amount of glucose to be imported into specific neural circuits, where it is used to form new memory-related proteins. Adler et al. discover that microglia, the resident immune cells of the brain, are critical for this process via a mechanism called microglial-metabolic coupling.

Mansouri et al. show that IRG1-derived itaconate is spatially depleted in lung tumors and that restoring it with octyl itaconate suppresses tumor growth. Mechanistically, itaconate inhibits G6PD activity and pentose phosphate pathway flux, reprograms macrophages toward anti-tumor states, and directly limits lung cancer cell proliferation.
