Mansouri et al. show that IRG1-derived itaconate is spatially depleted in lung tumors and that restoring it with octyl itaconate suppresses tumor growth. Mechanistically, itaconate inhibits G6PD activity and pentose phosphate pathway flux, reprograms macrophages toward anti-tumor states, and directly limits lung cancer cell proliferation.
