Viruses don’t just interact with cells biochemically — their mechanical properties also matter. We asked how modifying envelope proteins change stiffness and strength of HIV virus‑like particles (VLPs).

Using atomic force microscopy, we measured spring constant, breaking forces & deformation of VLPs.
Key result: VLPs containing influenza HA₂ sequences were mechanically stiffer and stronger than gp150 VLPs.

We produced three HIV VLP types with the same Gag protein shell but different envelopes:
• GagM + gp150
• GagM + gp140HA₂tr
• GagM + gp120HA₂
This allowed us to isolate the mechanical role of the envelope proteins.

Why it matters: VLP mechanics influence stability, entry, and vaccine performance. Envelope modifications may allow us to mechanically engineer VLPs, adding a new dimension to vaccine design.
