With an open access imaging pipeline from Roy Chowdhury @prudentlab.bsky.social, and modelling of heteroplasmy shifts and genetic bottlenecks by @abhilesh.bsky.social. All code freely available on github.com/JvdAlab/mtDN....

We can't distinguish single-nucleotide variants (yet), but do visualise mtDNA deletions and distinguish D. yakuba from D. melanogaster mtDNA at single-molecule level in situ, in whole-mount tissue and human cells. Do give it a try, and let us know how it works in your hands and model systems!

Wonderful work from our lab's first PhD student Rajini Chandrasegaram, with Antony Hynes-Allen, Stavroula Petridi, @beitongtong.bsky.social, @abhilesh.bsky.social. Thanks to great collaborations with Hansong Ma, @prudentlab.bsky.social @mitocamb.bsky.social @frisophagy.bsky.social!
A Research Briefing on our MitoPerturb-Seq paper just came online @natsmb.nature.com too: www.nature.com/articles/s41.... Hopefully even more accessible than the manuscript! 😉
Really pleased to see this out @natsmb.nature.com: www.nature.com/articles/s41.... MitoPerturb-Seq combines #singlecell #CRISPR screening with whole-cell Multiome, to identify nuclear modifiers of mtDNA dynamics. @mitocamb.bsky.social @mrc-mbu.bsky.social @prudentlab.bsky.social and Stephen Burr
