Excited that my short perspective on how public engagement has shaped my research is now out in @cp-immunity.bsky.social - I hope u enjoy, lots of inspiration to be had from all the stories #ScienceEngagement @exposomeimmunology.bsky.social @fbmh-uom.bsky.social @lydiabeckeriii.bsky.social

Specialized fibroblastic reticular cell (FRC) subsets in lymph nodes orchestrate the compartmentalization that guides immune cell migration, interaction, and function. Alouche et al. examine the signals governing FRC differentiation and identify a Notch2-dependent process by which homeostatic mature dendritic cells drive Ccl19hi T-zone fibroblast differentiation, maintain segregation of distinct T-zone niches, and support CD8+ T cell memory formation.

Interleukin-17-Producing γδ T Cells Originate from SOX13+ Progenitors that Are Independent of γδTCR Signaling
NK cell dysfunction and interferon-γ production underlie autoinflammation in mevalonate kinase deficiency
The mechanisms linking defective protein prenylation to systemic inflammation in mevalonate kinase deficiency remain unclear. Munoz et al. reveal that mice and humans with MKD have defective NK cells, with impaired granule trafficking, reduced cytotoxicity, and increased IFN-γ production. Their findings help explain the cause of inflammatory flares in MKD and provide a rationale for testing JAK inhibitor therapy.

Spatiotemporal analysis reveals distinct inflammatory programs underlying chronic colitis
Fransson, Sorini, et al. build a multi-model, longitudinal transcriptomic atlas of experimental chronic colitis, integrating bulk, single-cell, and spatial data. They reveal shared inflammatory programs, epithelial antigen presentation, and dynamic neutrophil states, linking mouse models to human IBD and highlighting epithelial-immune crosstalk as a therapeutic target.

TREX1, but not other DEDDh exonucleases, restrains cGAS-STING activation by cytosolic DNA. Zhu et al. examine the basis for this specificity and define a modular double-stranded DNA (dsDNA)-recognition architecture within TREX1 comprising R128, a substrate-sensing loop, and an auxiliary DNA-binding surface, linking dsDNA recognition to immune control and antitumor immunity.

MAIT cells acquire effector-memory and tissue-residency programs in the thymus. Paiva et al. demonstrate that MAIT cells can be selected by epithelial cells and egress the thymus at several developmental stages with subset-specific tissue tropisms and that during colitis, thymic MAIT17 cells expand and are directed to the gut.

Modular double-stranded DNA recognition defines specificity of the exonuclease TREX1 in cGAS-STING control
MAIT cells egress the thymus at several maturation stages with selective capacity to seed different tissues
Loss of B cell tolerance, autoantibody production, and immune complex deposition are hallmarks of systemic lupus erythematosus. Al Souz et al. find that kidney-infiltrating CD8+ T cells differentiate from stem-like precursors in renal lymph nodes and cause tissue injury in lupus nephritis, demonstrating that pathological organ-specific responses mediated by stem-like CD8+ T cells are embedded within systemic autoimmunity.

Organ injury in systemic autoimmunity is mediated by stem-like CD8+ T cells arising from tissue-draining lymph nodes
