Using the SARS-CoV-2 mRNA vaccine as a model immune challenge, Montamat-Garcia et al. describe how antibody class-switch recombination (CSR) during a primary human response is partially sequential, but with checkpoint at the IGHG2 locus. Moreover, CSR is influenced by the B cell subset and VDJ gene usage.
