HIV remains a health issue, with new therapies needed. Cromarty et al. show that HIV tri-specific killer engagers bind to and mediate effective NK cell functions against HIV-infected cells. In rhesus macaques, HIV TriKEs expand and activate the NK cell population, similar to other licensed IL-15-containing agents.
