⚡New preprint from the lab⚡: A key early checkpoint in gene expression is promoter proximal pausing of RNA polymerase II. For over 20 years, we as field have not been able to recreate pausing under realistic cellular conditions. @robertovn.bsky.social shows that ELOF1 is the missing piece.

This solves a major question in the field on how TFIIF (an initiation factor) is "resisted" by RNA polymerase II at pause sites. DSIF and NELF are not enough, but with ELOF1, TFIIF does not stand a chance. Read all about it here: tinyurl.com/ELOF1-pause
Using functional genomics, our new cell free genomics approach GATO-seq (tinyurl.com/gato-seq), structural biology, and biochemistry, we show that ELOF1 with its co-conspirators DSIF and NELF can induce promoter proximal pausing under physiological conditions.
