Preprint alert đšHappy to share my first Posdoc manuscript from the Vos lab @voslab.org. We introduce GATO-seq, a high-throughput, fully in vitro technique that maps RNA polymerases at base pair resolution over time. Using it, we found a new pause state of Pol II: sidetracking tinyurl.com/superpause
Please join Andreas Ladurner and me for the GRC Chromatin Structure and Function conference to be held near Barcelona Spain from May 31-June 5! We accepting abstracts for posters and talks. The meeting is preceded by an awesome GRS. Learn more in our video below and apply www.grc.org/chromatin-st...
âĄNew preprint from the labâĄ: A key early checkpoint in gene expression is promoter proximal pausing of RNA polymerase II. For over 20 years, we as field have not been able to recreate pausing under realistic cellular conditions. @robertovn.bsky.social shows that ELOF1 is the missing piece.
Big thanks to our lab and our collaborators from the lab of Andreas Mayer, Annkatrin Bressin and Zhihao Shao at
@molgen.mpg.de and the Institute of RNA Biology and Genomics at the Medical University of Innsbruck
This solves a major question in the field on how TFIIF (an initiation factor) is "resisted" by RNA polymerase II at pause sites. DSIF and NELF are not enough, but with ELOF1, TFIIF does not stand a chance. Read all about it here: tinyurl.com/ELOF1-pause
Using functional genomics, our new cell free genomics approach GATO-seq (tinyurl.com/gato-seq), structural biology, and biochemistry, we show that ELOF1 with its co-conspirators DSIF and NELF can induce promoter proximal pausing under physiological conditions.
Amazing work from Bonnie Su and Laura Caccianini from our group, the lab of @lucas.farnunglab.com with @mosorio91.bsky.social and Alex Stone and the lab of @andersshansen.bsky.social with Masahiro Nagano!
đšNew preprint! We built a cell-free genomics platform (GATO-seq) to probe transcriptional regulation and discovered a âsuper pauseâ sequence that triggers a new Pol II active-site conformation. Huge shout-out to @robertovn.bsky.social for pulling off this monster of a project. tinyurl.com/superpause
đ§Șđ§ŹNew preprint We present cryo-EM structures of reconstituted CTCFânucleosome complexes, showing CTCF dimerization drives nucleosome oligomerization into defined higher-order assemblies. Disrupting CTCFâCTCF interfaces in mESCs reduces looping and impairs differentiation. tinyurl.com/CTCF-nucleos...

The E3 ubiquitin ligase mechanism specifying target-directed microRNA degradation (TDMD) is now published! đđŸ We, @bartellab.bsky.social and Schulman lab, describe how 2-RNA factors control protein degradation by recruiting an E3 ligase. @mpibiochem.bsky.social

đš New preprint: GATO-seq did it again! ELOF1 is the missing promoter-proximal factor that confers RNA Pol II resistance to TFIIF. Including ELOF1, DSIF, and NELF in GATO-seq reactions recapitulates promoter-proximal pausing in vitro at physiological conditions for the 1st time.
