We extended this approach to alkylglycerols and sphingosines to selectively access subsets of ether lipids and sphingolipids
Systematic chemical modifications enabled new and more selective metabolic incorporation, allowing to target subsets of glycero(phospho)lipids.
Together, our study shows that lipid tail structure is a programmable determinant of metabolic fate and that this principle can be leveraged to selectively route substrates through endogenous lipid metabolic networks enabling selective editing and functionalization.
We started out by mapping the structural dependence of lipid metabolism using isotope tracing, showing that lipid tail structure directs metabolic fate across the lipidome (Figure 1)
Finally, we introduced bifunctional lipid probes that enable selective labeling and visualization of distinct lipid classes in living cells
