We subsequently developed a charge correction tool to turn improperly calculated charges (from multiple ions measured together, decay of the charge during data collection) into the correct charge value. This correction results in the Gaussian isotopic envelope typical of proteins.
This was a super fun time and it is always nice to talk about paleoproteomics.
We measured single/limited numbers of ions from intact proteins of various sizes including mixtures and applied the STORI slope calculation to calculate the charge.
