In this Marble, the authors report an attempt to monitor DNA methylation at major satellite repeats using a two-component system: a zinc-finger anchor targeting the satellite repeats, and an MBD detector targeting 5mC methylation marks.
While each component bound to its specific target as expected, co-transfection of both halves produced a strong signal even in the absence of DNA methylation, suggesting spontaneous complementation of the split-Venus halves rather than a true methylation readout.
