This unique genetic toolkit allowed us to that (1) oxidative inactivation by DAO1/DAO2 follows GH3‑mediated conjugation, defining the main IAA inactivation pathway hierarchy; (2) IAA glycosylation is more important than previously thought;
(3) the gh3oct mutant likely retains some GH3 function, helping explain prior inconsistencies; and (4) the IAA overproducing plants gh3oct and gh3oct dao1-6 plants show a premature downregulation of (my favorite!) FLOWERING LOCUS C likely underlying their early flowering phenotype
Aiming to understand IAA metabolism and the biological roles of the genes involved, Rubén and I generated as many mutant combinations as we could imagine. We then fed plants isotope labelled IAA and followed its metabolism thanks to the expertise of the Ondrej Novák´s team.
