Sun and Fisher performed chemical-genetic dissection of the conserved transcription elongation factor SPT5 in human cells, which revealed that CDK9 phosphorylates three distinct regions in SPT5 that coordinately regulate RNA polymerase II pausing, elongation rate, pre-mRNA splicing, and termination to ensure productive transcription and cell viability.
