Cryo-EM structures of human dopamine transporter (hDAT): apo, substrate-bound, & antidepressant-bound. Bupropion binds central pocket in both DAT & NET, explaining dual NRI/DRI action. Vanoxerine traps DAT in outward-open state (distinct pose). K+ occupies Na+ sites, suggesting regulatory role.
