We are able to create 3D movies of the exodermis barrier across developmental stages for both wheat and maize using this simultaneous triple staining method. Here is an example of maize root, where green indicates Auramine O staining (lignin + suberin).

I am very excited to share my simultaneous triple staining method for monocot roots. This method is not only faster but also utilizes a smaller volume of stain than traditional methods and minimizes mechanical tissue damage.

We tested our protocol with the roots of two crop plants (wheat and maize). Our images allow us to visualize and quantify cellular components across different cell types, tissues, and developmental stages.
