(3/5) We are able to create 3D movies of the exodermis barrier across developmental stages for both wheat and maize using this simultaneous triple staining method. Here is an example of maize root, where green indicates Auramine O staining (lignin + suberin).
(1/5) I am very excited to share my simultaneous triple staining method for monocot roots. This method is not only faster but also utilizes a smaller volume of stain than traditional methods and minimizes mechanical tissue damage. www.biorxiv.org/content/10.6...
(4/5) You can tune into the supplemental movies of our preprint to see more developmental regions. Here is an example of wheat root, where green indicates Auramine O staining (lignin + suberin). More videos on lab YouTube: www.youtube.com/@ArifAshraf
(5/5) I am grateful to my advisor @aribidopsis.bsky.social, Ashraf Lab members, and our collaborator @rajuaafc.bsky.social for constant support. It wouldn’t be possible without generous funding from NSERC and CFI; and the supportive @ubcbotany.bsky.social @science.ubc.ca grad program.
(2/5) We tested our protocol with the roots of two crop plants (wheat and maize). Our images allow us to visualize and quantify cellular components across different cell types, tissues, and developmental stages. Lab website (scroll at the end): www.ashraflab.com/what-we-dont...

🔬🍀🎉 I am happy and excited to share that graduate student, @ohazel.bsky.social Olivia's first author research article is published now @newphyt.bsky.social We are grateful to editor, reviewers, and @newphyt.bsky.social to help us to improve the story nph.onlinelibrary.wiley.com/doi/10.1111/...
🔬🍀🎉 Really excited to see that @science.ubc.ca UBC Research Excellence cluster highlighted the wheat image, captured by @juliazheku.bsky.social Julia, from our lab in the cover this year.
I am excited to share that Julia will start grad school @science.ubc.ca @ubcbotany.bsky.social with us from Fall 2025! Julia graduated from the University of Massachusetts Amherst and worked on root development with Tobias Baskin. She is my grad advisor's postdoc advisor's student!
🔬🔬🔬 Many of you have seen @juliazheku.bsky.social Julia's pretty wheat and maize root images. Here is an effort of growing root in the messy soil environment.

Our fantastic #FluorescenceFriday featured image was acquired by @juliazheku.bsky.social, a graduate student with @aribidopsis.bsky.social. The image shows a maize cross section, with a developing nodal root breaking through the basal stem node. Check out our post ⤵️:
